These mice were also genetically engineered to have large numbers of Th2 helper cells that recognize the same antigen in the context of MHCII. So instead of the normal situation in non-transgenic mice where there may be only a few dozen Th1 and a few dozen Th2 helper cells that recognize MOG92-106, there are now millions even before birth. In a sense it is as if the mouse even before birth has recognized and responded to MOG92-106 antigen, as if it has been vaccinated against MOG92-106. So it is a transgenic form of experimental allergenic encephalomyelitis (i.e., EAE) and many people question whether EAE is actually a model of MS.

So now we have a mouse line vaccinated against MOG92-106. In one institution maybe 35% of these mice develop a disease that resembles remitting/relapsing MS while in another facility up to 90% of the mice develop remitting/relapsing disease. The authors hypothesized that the differences between research facilities might be explained by differences in gut flora in the mice of different facilities.

The authors show that if the mice are raised from birth in a germ-free facility where there were no bacteria in their gut or in their environment they did not develop disease. Germ-free mice whose guts were repopulated with bacteria all developed the remitting/relapsing disease.

The explanation for these findings is likely that some gut bacteria have an antigen that molecularly resembles MOG92-106. The Th1 cells encountering and recognizing this gut bacterial antigen in the context of MHCI will become activated and proliferate and help activate and cause to proliferate those CD8+ cells that also recognize the MOG92-106 mimetic causing these cells to mount an attack against cells presenting MOG92-106 in the context of MHCI (and this could only be oligodendrocytes since only these cells synthesize MOG). Furthermore, Th2 cells encountering this gut bacterial mimetic of MOG92-106 will also proliferate and activate the B cells that specifically recognize the bacterial mimetic of MOG92-106 causing these cells to produce lots of antibodies directed against MOG92-106.

[B]Thoughts[/B]: Yes, our immune system is complex and no doubt molecular mimicry can play a role in autoimmune diseases and our gut flora may well be one of many contributing factors to MS, or in some individuals with MS. But we need our gut bacteria for health. Because of the variation from person to person in MHCI, MHCII and TCR composition it is unlikely we can find one common bacterial mimetic molecule that may play a role. Factor A may be the mimetic in person A, while Factor B in person B. etc, etc. It may well be that if gut bacteria is a contributing factor (likely one of many contributing factors), we will never be able to identify the one mimetic responsible for being the contributory factor. Ik heb gekeken of ik onderzoeken kon vinden waar nader in gegaan wordt op probiotica.

Omdat MS een ontstekingsziekte is, zou bij MS het eerste ongunstig zijn en het tweede gunstig. Op dit moment is nader onderzoek nodig om te bepalen of de probiotica het afweersysteem in een voor MS gunstige of ongunstige richting sturen.[/QUOTE]

Iedereen kan in 3 typen darmflora (enterotypes) ingedeeld worden.
Interessant is dat er genoemd wordt dat de darmflora vanaf een jaar of 4 ongeveer hetzelfde blijft. Verandering in voeding kan het wel wijzigen, maar dat gaat langzaam. Met een faecal transplant kan de samenstelling van de darmflora ingrijpender en sneller gewijzigd worden.

Ja Marsei, ik denk dat de darmen DE hoofdoorzaak van vele ziekten - en dus ook m.s.- zijn. Daar begint het. Alleen het duurt wel even (paar jaar) voordat de darmen (o.a. via "goede" voeding en probiotica) verbeterd zijn.
Vanuit die slechte darmen komen al die symptomen -waaronder ccsvi- voort.

De genetische factor speelt ook een rol, CCSVI is ook deels aangeboren.
Ik heb anderhalf jaar een superstreng dieet gevolgd, geen rund, geen zuivel, geen gluten etc.....geen resultaat.
Dus of bij mij de darmen een rol spelen, ik geloof het niet...

Off topic, maar.... complimenten wederom voor Marsei. Zoveel wetenschappelijke informatie zo snel bij elkaar gezet. Ik vind het indrukwekkend.
